my MC have high perception so he can see what side she drink it

Or it may be that Melissa throws the water for two reasons / reactions:
1-if the MC is a jerk with sofia she then to tell him to Melissa that he was a idiot (or something more insulting) and Melissa was angry because she does not believe that
2 - but if the MC is good with her Melissa would be a bit jealous and sofia could have told him (sarcastically) that the MC was dying for her or something like that and that active her yandere side
